Oligonucleotide Analysis and Purification
Depending on the synthetic methodology used, oligonucleotides can be purified many ways including:
· Reverse Phase LC
· Ion-exchange LC
· Gel-filtration Chromatography
· Ultra-filtration
Azco supplies AMBERCHROM™ and AMBERLITE™ brand polymeric reversed phase and ion exchange resins from Rohm and Haas and also supplies OLIGOSEP™ and OLIGOPREP™ brand polymeric reversed phase columns from Transgenomic. All of these materials have been optimized for the analysis and purification of oligonucleotides and are readily scalable to meet any peptide requirement from the bench to large scale manufacturing. Rohm and Haas purification products are manufactured under GMP conditions with all necessary supporting regulatory .documentation.
AMBERCHROM™ resins are highly efficient materials for reversed phase chromatography without the limitations of silica. These resins are designed for the purification of proteins, peptides and oligonucleotides. The OLIGOSep/Prep™ Family
There are two products manufactured by Transgeomic, specifically for the "high-speed", ion-pair, revesed separation of oligonucleotides. They are:
· OLIGOSep™ is a 2 micron, fully alkylated, C18 polymeric reversed phase column. This material is designed to withstand pH in the range of 0 to 14 and heating up to 100C. Their small particle size gives them excellent efficiency for reversed phase separation, and the pH and temperagure ranges provide dimensions for separation optimization.
· OLIGOPrep™ columns offer the same benefits as above; however, they use 5 micron particle size resins to allow for higher flow rates; thus, faster purification of oligonucletides.
The Amberchrom™ Family
· AMBERCHROM™ XT polishing grade resins demonstrate high efficiency with improved selectivity for difficult separations when compared to silica RPC resins. They offer high capacity and high flow rates in medium to high pressure production applications.
· AMBERCHROM™ CG chromatography grade resins. These materials have been commercially proven for many years, AMBERCHROM™ CG resins are suitable for use in many peptide and oligonucleotide applications including, front end capture, purification, flow-through, and desalting depending on the pore size and particle size selected.
· AMBERLITE™resins are low cost, low efficiency, large particle size resins designed for capture or flow-through applications or any application where a low-cost adsorbent is required.
Reversed Phase Chromatographic Resins for Bimolecular Purification and Polishing
AMBERCHROM™ XT chromatographic media are a refinement of the existing AMBERCHROM™ CG line offering higher efficiency and higher pressure capabilities. ,Iimproved resolution is provided from the smaller particle sizes. The XT line is still pH stable over the entire range and provides the same stability to caustic CIP procedures. Theyse materials are targeted for high efficiency oligonucleotide purification whileproviding an economical and rugged alternative to silica for reversed phase chromatographic protocols.
AMBERCHROM™ XT media have demonstrated exceptional capabilities for purifying both naked as well as highly modified oligonucleotides and RNA. They are ideally suited for therapeutic oligonucleotides such as antisense or siRNA.
Downstream purification can be optimized by taking advantage of Amberchrom’s excellent pH stability in purifying a wide range of peptides and oligonucleotides. The resolving power of AMBERCHROM™ XT delivers high yield-purity in the separation, and their superior rigidity permits use with medium pressure equipment, up to 60 bar.
Available in two different particle size ranges (20 and 30 microns), AMBERCHROM™ XT resins are compatible with most organic solvents, dilute acids and bases.
Amberchrom™ CG Resins for High Performance Polymeric RPC Chromatography
AMBERCHROM™ resins provide the purification power for reversed phase chromatography without the limitations of silica media such as pH and temperature. Amberchrom resins are pH stable from 0 to 14 and are compatible with high temperature applications.
AMBERCHROM™ CG media are macroporous, polymeric reversed phase liquid chromatography resins. They are designed for laboratory and process scale purifications of proteins, peptides, nucleic acids, antibiotics, and small molecular weight pharmaceuticals.
Large surface area, unique pore size and pore volume distribution make AMBERCHROM™ CG300 ideally suited for the separation of peptides. This resin has high capacity for many pharmaceutical compounds and has been commercially proven for many years. Amberchrom is an excellent technical and economical alternative to RPC silica when used in high resolution, low pressure chromatography.
AMBERCHROM™ CG resins are available in three different particle size ranges: small (S), medium (M) and coarse(C), with, 35, 75, and 120 micron mean particle sizes respectively. These materials are supplied ready to pack as a slurry in 20 % ethanol. AMBERCHROM™ CG resins are ideally suited for operation within the entire pH range, and can be easily cleaned in place (CIP) with 1M Sodium Hydroxide. Amberchrom is compatible with most organic solvents and dilute acids and bases.
AmberLite™ Resins as Low Cost Adsorbants
AMBERLite™ resins are low cost polymeric resins designed for capture applications or those applications that call for an inexpensive adsorbant. Amberlite resins are also pH stable an can be easily cleaned. They are offered in the same pore sizes as the Amberchrom CG resins, but much larger particle sizes. These materials are ideal for fast flow capture or flow-through applications.
